In der vorliegenden Arbeit wurde der genetische Code des humanpathogenen Pilzes Candida albicans durch gezielte Etablierung und Optimierung eines orthogonalen Pärchens bestehend aus tRNA und tRNA-Synthetase um die synthetische Aminosäure p-Azidophenylalanin erweitert. Nach Integration der synthetischen Aminosäure in vivo und UV-Bestrahlung war eine positionsspezifische Kreuzvernetzung binärer Protein-Protein-Interaktionen möglich, wie anhand von zwei Modellproteinen gezeigt werden konnte. Diese kreuzvernetzten Proteinkomplexe konnten zudem aufgereinigt und massenspektrometrisch analysiert warden. Somit konnte die generelle Eignung der Methode für die Identifizierung auch unbekannter Interaktionen gezeigt werden und ein wirkungsvolles Werkzeug für die Candida-Forschung etabliert werden.
